An Absolute M E T H O D for Assay of Virus Hemagglutinins

چکیده

The ability of certain viruses to agglutinate red cells (1) is widely employed as a basis of titration, because of the speed and simplicity of the reaction. Usefulness of the method has been limited, however, by lack of a reliable means of translating hemagglutinating activity into an absolute number of active particles per cubic centimeter, so tha t values obtained for a single virus can only be expressed in relation to that of some standard suspension, and different viruses cannot be compared at all. Moreover the relationship between hemaggintination and infectiousness is often complex, wide variations in the ratios of these activities being produced by various t reatment of virus suspensions (2). An absolute method for determination of the number of hemagglutinating particles would materially increase the usefulness of the virus assay when the two activities are closely associated, and make possible precise analysis of the relationship between infectivity and agglutination in cases in which they are separable. Such a method is described in this communication.

Simultaneous Detection of Arabis Mosaic Virus, Cherry Leafroll Virus and Cucumber Mosaic Virus with Coamplification of Plant mRNA as Internal Control for Olive Certification Programs

Background and Aims: Certification programs of plant propagating materials rely on faster, cheaper and more importantly sensitive and reliable methods for detection of systemic pathogens as indicated in national and/or international health standards of plant propagating materials. Reverse transcription-polymerase chain reaction (RT-PCR) has been documented as an alternative assay for certific...

Development of qRT-PCR Test for Quantification of Rubella Virus in Commercially Available Vaccines

vaccines is considered as a critical point and in-process of quality control (IPQC) test of vaccine production. Rapid tests, like real time PCR, are more appropriated when the production occurs at industrial scale because of the amounts of starting materials and the excess of consumed time required. In the current study, a real-time quantitative reverse transcription-polymerase chain reaction...

Development of a Multiplex RT-PCR Assay for Detection of the Causal Agents of Citrus Tristeza and Cachexia Diseases with Coamplification of Plant mRNA as an Internal Control

Background and Aims: Plant certification programs need reliable, fast, cheap and sensitive methods for detection of systemic pathogens with special interest in virus and viroid detection. Reverse transcriptase-polymerase chain reaction (RT-PCR) has been documented as an alternative assay for certification of plant propagating materials. The main object of the present study was the optimization ...
